Genetic variations associated with susceptibility to bacteria linked to stomach disorders

May 7, 2013 ? Two genome-wide association studies and a subsequent meta-analysis have found that certain genetic variations are associated with susceptibility to Helicobacter pylori, a bacteria that is a major cause of gastritis and stomach ulcers and is linked to stomach cancer, findings that may help explain some of the observed variation in individual risk for H pylori infection, according to a study in the May 8 issue of JAMA.

"[H pylori] is the major cause of gastritis (80 percent) and gastroduodenal ulcer disease (15 percent-20 percent) and the only bacterial pathogen believed to cause cancer," according to background information in the article. "H pylori prevalence is as high as 90 percent in some developing countries but 10 percent of a given population is never colonized, regardless of exposure. Genetic factors are hypothesized to confer H pylori susceptibility."

Julia Mayerle, M.D., of University Medicine Greifswald, Greifswald, Germany, and colleagues conducted a study to identify genetic loci associated with H pylori seroprevalence. Two independent genome-wide association studies (GWASs) and a subsequent meta-analysis were conducted for anti-H pylori immunoglobulin G (IgG) serology in the Study of Health in Pomerania (SHIP) (recruitment, 1997-2001 [n =3,830]) as well as the Rotterdam Study (RS-I) (recruitment, 1990-1993) and RS-II (recruitment, 2000-2001 [n=7,108]) populations. Whole-blood RNA gene expression profiles were analyzed in RS-III (recruitment, 2006-2008 [n = 762]) and SHIP-TREND (recruitment, 2008-2012 [n=991]), and fecal H pylori antigen in SHIP-TREND (n=961).

Of 10,938 participants, 6,160 (56.3 percent) were seropositive for H pylori. GWAS meta-analysis identified an association between the gene TLR1 and H pylori seroprevalence, "a finding that requires replication in non-white populations," the authors write.

"At this time, the clinical implications of the current findings are unknown. Based on these data, genetic testing to evaluate H pylori susceptibility outside of research projects would be premature."

"If confirmed, genetic variations in TLR1 may help explain some of the observed variation in individual risk for H pylori infection," the researchers conclude.

Editorial: Helicobacter pylori Susceptibility in the GWAS Era

In an accompanying editorial, Emad M. El-Omar, M.D., of Aberdeen University, Aberdeen, United Kingdom, writes that the authors of this study are appropriate to state, "based on their data, genetic testing to evaluate H pylori susceptibility is premature."

"This would be superfluous, because nongenetic testing for the infection can be accomplished at a fraction of the cost. There is a bigger picture: understanding genetic susceptibility to H pylori is essential for understanding how to overcome this infection. The current approach to eradication of the infection is limited and based entirely on prescribing a cocktail of antibiotics with an acid inhibitor to symptomatic individuals. However, H pylori antibiotic resistance is increasing steadily, and eventually curing even benign conditions such as peptic ulcer disease arising from H pylori will be difficult. When considering gastric cancer, another H pylori-induced global killer, the necessity for understanding the pathogenesis of the infection and the role of host genetics in susceptibility is even greater. The corollary is that better understanding of infections, including genetic epidemiology, is crucial to design measures to eradicate the downstream consequences of H pylori in large populations."

More hurricanes for Hawaii?

May 5, 2013 ? News of a hurricane threat sends our hearts racing, glues us to the Internet for updates, and makes us rush to the store to stock up on staples. Hawaii, fortunately, has been largely free from these violent storms in the recent past, only two having made landfall in more than 30 years.

Now a study headed by a team of scientists at the International Pacific Research Center, University of Hawaii at Manoa, shows that Hawaii could see a two-to-three-fold increase in tropical cyclones by the last quarter of this century. The study, which appears in the May 5, 2013, online issue of Nature Climate Change, though, leaves open the question, how worried Island residents should get.

"Computer models run with global warming scenarios generally project a decrease in tropical cyclones worldwide. This, though, may not be what will happen with local communities," says lead author Hiroyuki Murakami.

To determine whether tropical cyclones will become more frequent in Hawaii with climate change, Murakami and climate expert Bin Wang at the Meteorology Department, University of Hawaii at Manoa, joined forces with Akio Kitoh at the Meteorological Research Institute and the University of Tsukuba in Japan. The scientists compared in a state-of-the-art, high-resolution global climate model the recent history of tropical cyclones in the North Pacific with a future (2075-2099) scenario, under which greenhouse gas emissions continue to rise, resulting in temperatures about 2?C higher than today.

"In our study, we looked at all tropical cyclones, which range in intensity from tropical storms to full-blown category 5 hurricanes. From 1979 to 2003, both observational records and our model document that only every four years on average did a tropical cyclone come near Hawaii. Our projections for the end of this century show a two-to-three-fold increase for this region," explains Murakami.

The main factors responsible for the increase are changes in the large-scale moisture conditions, the flow patterns in the wind, and in surface temperature patterns stemming from global warming.

Most hurricanes that might threaten Hawaii now are born in the eastern Pacific, south of the Baja California Peninsula. From June through November the ingredients there are just right for tropical cyclone formation, with warm ocean temperatures, lots of moisture, and weak vertical wind shear. But during the storms' long journey across the 3000 miles to Hawaii, they usually fizzle out due to dry conditions over the subtropical central Pacific and the wind shear from the westerly subtropical jet.

Surprisingly, even though fewer tropical cyclones will form in the eastern Pacific in Murakami's future scenario, we can expect more of them to make their way to Hawaii.

The upper-level westerly subtropical jet will move poleward so that the mean steering flow becomes easterly. Thus, storms from Baja California are much more likely to make it to Hawaii. Furthermore, since the climate models also project that the equatorial central Pacific will heat up, conditions may become more favorable for hurricane formation in the open ocean to the south or southeast of Hawaii.

"Our finding that more tropical cyclones will approach Hawaii as Earth continues to warm is fairly robust because we ran our experiments with different model versions and under varying conditions. The yearly number we project, however, still remains very low," reassures study co-author Wang.

First Person: Dismal U.S. Job Market Led an Iraq War Vet to Pursue His Passion

President Obama and Michelle Obama are pushing companies to hire more U.S. military veterans. They said Tuesday that, through a program called Joining Forces, employers have hired or trained 290,000 veterans since August 2011. Through short stories and photos, Yahoo News asked veterans whether they've found work since the start of the 2007 recession or, if not, how their job hunt is going.

FIRST PERSON | When I returned from Iraq in 2009, after having served as a machine gunner for a convoy security team in Mosul as a member of the Washington Army National Guard, I quickly found that the U.S. job market had dried up in my absence.

I'd been a licensed securities dealer before enlisting, but my securities license had expired while I was in Iraq, and none of the Wall Street firms that I spoke with cared to help me renew them, in spite of the fact that they'd lapsed due to my decision to volunteer for service during a time of war.

With seemingly few options available, and with the encouragement of some of my military councilors and closest friends who knew where my heart truly lay, I decided to take the plunge and write full-time.

I've moved to the Philippines to do so, and I am actually earning enough from my writing now to feed my family. What I thought was a curse, a very sour U.S. job market, has been my biggest professional blessing. I no longer do what I'm qualified to do. I now do what I love to do, and I've never been happier.

Researchers discover new target for personalized cancer therapy

A common cancer pathway causing tumor growth is now being targeted by a number of new cancer drugs and shows promising results. A team of researchers at Case Western Reserve University School of Medicine have developed a novel method to disrupt this growth signaling pathway, with findings that suggest a new treatment for breast, colon, melanoma and other cancers.

The research team has pinpointed the cancer abnormality to a mutation in a gene called PIK3CA that results in a mutant protein, which may be an early cancer switch. By disrupting the mutated signaling pathway, the Case Western Reserve team, led by John Wang, PhD, inhibited the growth of cancer cells, opening the possibility to new cancer therapies.

Their findings, "Gain of interaction with IRS1 by p110? helical domain mutants is crucial for their oncogenic functions," was published on May 2 in the journal Cancer Cell.

Cancer arises from a single cell, which has mutated in a small number of genes because of random errors in the DNA replication process. These mutations play key roles in carcinogenesis.

"This discovery has a broad impact on the treatment of human cancer patients because so many cancers are affected by this particular mutation in the p110? protein, which is encoded by the PIK3CA gene," said Wang, an associate professor in the Department of Genetics and Case Comprehensive Cancer Center. "This is a significant advance because we can now disrupt this misdirected signaling pathway in cancer cells."

"If you turn on a light, you have to turn on a switch. But in the case of the mutation of this protein, p110? turns on by itself," Wang said. "The mutation rewires the circuit and is uncontrolled. This implies that if you break these wires, you can control the growth of cancer. Our current discovery may lead to finding less toxic drugs that can be used for personalized treatment for cancer patients in the future."

"This research will impact the field by focusing us on new targets for treating and preventing metastasis in patients in a many different types of human cancers," said Stanton Gerson, MD, Asa and Patricia Shiverick-Jane Shiverick (Tripp) Professor of Hematological Oncology, and director of Case Comprehensive Cancer Center and of Seidman Cancer Center at University Hospitals Case Medical Center.

Wang's multidisciplinary team of Case Western Reserve researchers includes: Yujun Hao, Chao Wang, Bo Cao, Brett M. Hirsch, Jing Song, Sanford D. Markowitz, Rob M. Ewing, David Sedwick, Lili Liu and Weiping Zheng.

Sun erupts with superheated plasma

The sun fired off super-hot plasma in a dazzling eruption, captured by NASA's Solar Dynamics Observatory.

A coronal mass ejection (CME) erupted from just around the edge of the sun on May 1, 2013, in a gigantic rolling wave. CMEs can shoot over a billion tons of particles into space at over a million miles per hour. This CME occurred on the sun's limb and is not headed toward Earth. The video, taken in extreme ultraviolet light by NASA's Solar Dynamics Observatory (SDO), covers about two and a half hours.

Superheated plasma bursted from the sun's edge on May 1 in a gigantic rolling wave, said NASA, which published a?dazzling video?captured by its sun-watching spacecraft.

This solar eruption is known as a coronal mass ejection, or?CME, a kind of solar storm on the sun's outer atmosphere?that can send electrically charged particles into space. When a CME explodes in our direction, it usually takes three days to arrive. CMEs can occur simultaneously with solar?flares, another kind of sun storm involved in releasing energy associated with sunspots.

A large CME can discharge?a billion tons of particles into space at several million miles per hour.?Solar matter gushes out through the interplanetary medium, hitting any planet or spacecraft along its way.?

Like solar flares, CMEs, when directed at the Earth, can generate a space weather event called a geomagnetic storm, which occurs when they interact with the Earth's magnetic envelope for an extended period of time. CMEs at this intensity are unlikely to disrupt electrical systems on the Earth or interfere with GPS or satellite-based communications systems. But they may cause auroras near the poles.

The most powerful Earth-directed CMEs can also imperil astronauts and satellites in?orbit, and they can even damage ground-based power infrastructure.

The latest CME, however, took?place on the edge of the sun and didn't point at the Earth, said agency officials. The video,?taken in extreme ultraviolet light by NASA?s Solar Dynamics Observatory (SDO), shows that the sun's surface emitted a shimmering wave of plasma that splashed into open space. The event lasted two and a half hours.

Launched in 2010, the $850 million SDO satellite has provided detailed views of solar flares, CMEs, and other space weather events?in different wavelengths, including the extreme ultraviolet range of the light spectrum used to make the May 1 solar eruption video.

Wednesday's solar explosion was another indication that the sun is reaching the peak of its cycle, which lasts about 11 years. On April 12, the SDO observed an Earth-directed CME, preceded by?the most energetic?solar flare of the year.?

The National Oceanic and Atmospheric Administration experts have predicted that the solar maximum is likely to?occur this May.?

"But what we?ve come to find is the activity has really toned down over the last couple months," said Alex Young at NASA's?Goddard?Space Flight Center?in a NASA's podcast?on Wednesday. "We reached really the peak towards the end of 2011."

Because many of the previous solar cycles actually had two peaks, he believed that another solar maximum is going to happen.

"We?ve reached one of those humps and think that eventually the activity will pick back up and we?ll see another hump, a double hump solar maximum," Young said.

Do scientists expect a huge solar storm in 2013? On its website, NASA says that?no current observations or data?indicate an imminent solar?catastrophe. Scientists believe the intensity of the upcoming solar maximum will be similar to the previous maximum in 2002.
